Test Description and clinical significance

Genes
U2AF1

Disease:
Myelodysplastic neoplasm (MDS), chronic lymphocytic leukemia (CLL), Acute myeloid leukemia (AML), chronic myelomonocytic leukemia (CMML), lung adenocarcinoma, colon adenocarcinoma, and endometrial endometrioid adenocarcinoma

Next generation sequencing of gene panel. U2AF1 is one of several spliceosome complex genes frequently mutated in a variety of hematologic malignancies, particularly de novo myelodysplastic neoplasms (MDS), as well as lung and pancreatic cancers. Two hotspot mutations (S34 and Q157) occur within the two zinc-finger domains of the U2AF1 protein. These mutations have been associated with altered splicing patterns in vitro and in vivo.

Specimen Requirements

Collection:
At least 0.5 ml EDTA, citrate (ACD) or Heparinized whole blood or bone marrow aspirates for DNA mutations

Stability:
96hours/4 to 7 days refrigerated

Unacceptable Conditions:
Specimen submitted is in the incorrect fixative or anticoagulant. Specimen received after the stability date. Gross hemolysis. Frozen specimen. Insufficient specimen. Unlabeled. Left un-refrigerated for an extended period.

Storage & Transport

Blood, bone marrow specimens should be kept at room temperature and preferably received within 96 hrs of drawing. Samples that are 4 to 7 days old should be refrigerated.
